Gameplay in Grand Theft Auto V is an open-world action-adventure game played from a third-person or first-person perspective (only for newer versions of Xbox One, PlayStation 4, and PC). Players complete multiple missions - linear scenarios with predetermined objectives - to progress through the story. Outside of missions, players are free to explore the game world, which is fully present from the start, although certain gameplay concepts are unlocked as the story progresses.

Players can use melee attacks, firearms, or explosives to fight enemies, and run, jump, swim, or drive a wide variety of vehicles to explore the game world. Due to the very large size of the map, the game introduces the ability to fly planes, absent from all previous titles in the series, with the exception of Grand Theft Auto: San Andreas. In combat, the player can use an auto-aim system or cover to make fights easier. If the player is injured, their health level will begin to decrease, but it regenerates back to half if they are no longer injured. When the player dies or is "wasted", he will respawn at the nearest hospital with all weapons and health level again full, but he will be out of money. If the player commits crimes, then they will attract the attention of the authorities, marked by a "wanted" meter at the top of the HUD, which can be up to a maximum of 5 stars (in previous games in the series, the maximum number of stars possible was 6) . The higher the number of stars, the harder the authorities will try to kill or arrest the player, and at high stars, even the FBI and SWAT will join the chase. To escape the authorities, the player must exit their search area, highlighted on the mini-map at the bottom of the HUD.
